Investigation of biomolecules using mass spectrometry based methods

Abstract : Mass spectrometry (MS) has become a powerful tool of investigation in many fields of studies such as molecular physics, analytical chemistry and proteomics. The fast and sensitive performance of MS places it among the leading techniques in drug analysis and development. Furthermore, MS represents a promising approach to investigate structure and properties of biomolecules. Commonly used fragmentation techniques for peptide sequencing and recognition cannot provide fragmentation patterns for full characterization of selected precursor. They usually complement each other providing additional information about investigated polypeptide. The possibility of activating electrospray ionization produced polypeptide ions using a high-energy beam of positively charged air ions was investigated at the Physiological chemistry division, Karolinska Institutet. Obtained results will be described in this talk. The second part of presentation concerns the recent results of the project focused on the investigation of fundamental principles of chiral recognition and selection. Although tandem mass spectrometry has proven to be a highly promising method of quantitative chiral analysis, almost nothing is known concerning fundamental physical processes which lie behind gas-phase chiral recognition via activated fragmentation. Collisions activated dissociation of L- and D-forms of protonated amino acids with non-chiral and chiral targets were studied using MS based method on the ion source platform at the Department of Physics, Stockholm University.
